Applications are invited for a Research Assistant position to work on the molecular and metabolic basis of inflammation in Anti-Neutrophil Cytoplasmic Antibody (ANCA)-associated vasculitides (AAV) based in Trinity Translational Medicine Institute (TTMI), Trinity College Dublin. Autoimmunity results from a breakdown in the natural processes of self-tolerance resulting in the aberrant production of autoantibodies directed against our own proteins with a wide range of molecular consequences that underlie the pathology of autoimmune disease. In AAV, these autoantibodies are typically directed against two proteins, PR3 and MPO that are present on the surface of two major immune cell types: neutrophils and monocytes. When these antibodies bind their target proteins on the surface of neutrophils, they drive activation of these cells resulting in acute, local inflammation of vascular walls, most commonly in the respiratory tract and kidneys.
The current Vasculitis UK funded translational research project will explore the role that glycosylation of the Fc portion of the IgG molecule plays in driving relapse of AAV. The work leverages off a large bioresource (The RIV registry and biobank), which provides access to biosamples and clinical data from >700 patients with AAV. The goal of the project is to develop prognostic biomarkers that stratify patients with AAV according to relapse propensity, thus facilitating personalisation of immunotherapy.
